Product Introduction

FrCas9 Nuclease is a novel type II-A CRISPR-Cas9 system derived from Faecalibaculum rodentium, adding an important new member to the genome editing toolbox. This protein consists of 1372 amino acids, with its most distinctive feature being its unique palindromic PAM recognition sequence, 5'-NNTA-3'. This concise dinucleotide core PAM not only significantly expands the genomic targeting range but also endows FrCas9 with a special capability: the ability to directly target the TATA-box region within eukaryotic promoters. As a key element of core promoters involved in regulating transcription initiation, the TATA-box makes this characteristic a novel tool for studying TATA-box-related diseases and potential therapeutic interventions.

While maintaining editing efficiency comparable to, or even higher than, SpCas9, FrCas9 exhibits significantly lower off-target effects. Assessed by high-throughput technologies such as GUIDE-seq and AID-seq, the number of off-target sites for FrCas9 is far fewer than for SpCas9, and it shows lower tolerance for mismatches, demonstrating stricter recognition specificity.
